Constraint regions (or Rules by area) allow users to set different physical, spacing and same net spacing rules based on an area of the PCB. Example, this might be a flexi rigid where the track thickness should increase to improve manufacturing processes or a BGA where the default rules on the PCB might be too large to physically breakout the BGA. The basic principle is to draw an area (shape) where such rules are required, define the layers of the PCB you need the region for and then define rule sets for such areas.
